以硅烷醇C透皮研究指导配方设计
Design Product Formulation by Studying Percutaneous Penetration of Siloxanetriol Alginate
【摘要】 利用体外经皮渗透和脂肪组织体外培养技术 ,探讨了硅烷醇C经皮渗透和配方的促渗透 ,以及对脂肪组织的脂解作用。结果表明 ,硅烷醇C浓度在 1 0 %~ 50 % ,可呈不同程度地经完整的小鼠皮肤渗透 ,有剂量 反应关系。配方的组分对硅烷醇C经皮渗透有明显的影响 ,尤其是促渗透剂的应用。含芳香精油与氮酮复合的配方 ,促硅烷醇C在 2 4h的皮肤渗透量是对照配方的 3倍。硅烷醇C在体外有明显的促脂肪组织脂解作用 ,受试终浓度为 0 2 5mg/mL ,体外脂肪组织甘油释放量为 0 55μmol/g~ 0 86 μmol/g组织 ,生理盐水 (对照 )为 0 1 6。研究结果为配方设计提供了依据。
【Abstract】 Effect of percutaneous penetration,including influence of different formulations on penetration,and lipolytic activity of Siloxanetriol alginate were studied using the techniques of percutaneous penetration in vutro and adipose tissue incubation in vitro.The results showed that Siloxanetriol alginate ( 10?% ~ 50?% )penetrates through excised intact mouse skin with dose-response relationship,and has an obvious lipolytic activity on adipose tissue in vitro.The glycerol release from adipose tissue incubated with Siloxanetriol alginate is 0 55?μmol/g tissue at the concentration of 1?mg/mL ,compared with 0 16 of the control(saline).The penetration property of Siloxanetriol alginate greatly influenced by components of the formulation,especially in application of permeation enhancers.The cumulative penetration rate at 24?h of Siloxanetriol alginate in the formulation containing Azone and aromatic increases by two times,compared with the control formulation without penetration enhancers.
