引物原位标记技术在鼻咽组织印片中的应用

【摘要】 目的:探讨引物原位标记技术应用于鼻咽组织印片中的可行性。方法:以鼻咽非癌组织和鼻咽癌组织印片为研究材料,外周血正常淋巴细胞分裂中期染色体作对照,采用7号染色体特异性引物和引物介导原位DNA合成技术检测鼻咽印片组织细胞染色体数目。结果:6例鼻咽印片组织细胞7号染色体都获得特异性标记,3例正常组织的标记率为99.0%,二倍体细胞达94.0%;3例鼻咽癌细胞标记率为97.7%,二倍体细胞只有70.7%,二者差异显著(P<0.05)。结论:引物原位标记技术可用于组织印片中染色体的检测,初步研究结果提示7号染色体数目的改变可作为判断鼻咽恶性肿瘤的重要参考指标。

【Abstract】 Objective: To determine the feasibility for applying primed in situ (PRINS) labeling in the touch preparation of nasopharyngeal tissues. Method: Chromosome 7 in the touch preparations of six nasopharyngeal tissues were detected using primed in situ DNA synthesis and chromosome 7 specific primer. Results: Chromosome 7 in all the touch preparations were specifically labeled comparing with metaphase chromosome of peripheral blood lymphocyte. In three normal nasopharyngeal tissue and three nasopharyngeal carcinoma, labeling rates of chromosome 7 were 99% and 97.7%,respectively, and diploid cells were 94% and 70.7%, respectively. There was a significant difference between tumor and non tumor (P< 0.05). Conclusion: The results indicated that PRINS method can be used to detect chromosome aberrations in touch preparation and the number change of chromosome 7 might be an important target for diagnosing malignant tumor of nasopharynx.
