一氧化氮合酶在乳腺癌中的表达及其与血管形成机制的探讨
Expression of Inducible Nitric Oxide Synthase in Breast Cancer and Its Relation to Carcinoangiogenesis
【摘要】 目的:研究诱导型一氧化氮合酶(inducible nitric oxide synthase,iNOS)在乳腺癌中表达及与乳腺癌血管形成和淋巴结转移的关系。方法:采用免疫组化SP法检测60例乳腺癌中iNOS和血管内皮生长因子(vascular endothelial growth factor,VEGF)的表达,同时检测微血管密度(microvessel density,MVD,以CD34标记)。结果:iNOS阳性表达率为71.67%,在淋巴结阴性组和阳性组分别为53.3%和90%,两组间差异有统计学意义(P< 0.01);在iNOS阴性组CD34标记均值为13.100,而iNOS阳性表达(+)、(++)、(+++)组中的CD34标记均值分别为20.846、22.562、23.717,iNOS阴性和阳性组CD34均值差异有显著统计学意义(P< 0.01)。而iONS表达与VEGF表达则无相关性。结论:iNOS在乳腺癌中表达与肿瘤血管形成、淋巴结转移密切相关;但它可能不参与VEGF诱导血管生成的作用,而通过另外途径产生效应。
【Abstract】 Objective:This study was designed to investigate the expression of inducible NOS (iNOS) in breast cancer and its relation to angiogenesis in breast neoplasm and lymphocyte metastasis. Methods:The expression of iNOS and vascular endothelial growth factor (VEGF) in 60 patients with breast cancer were evaluated by immunohistochemistry SP method with anti-iNOS and anti-VEGF as the labels. Microvessel density (MVD) of breast cancer was also determined with anti-CD34 as the label. Results:The expression rate of iNOS in 60 cases was 71.67%, and the rate of node negative and node positive cancer were 53.3%and 90%respectively, with significantly different (P< 0.01). There was a significant difference of MVD (P< 0.01)but no statistical difference of expression of VEGF among groups of different iNOS expression. Conclusion:The expression of iNOS in breast cancer was high, and it had close relation to carcinoangiogenesis and metastasis. Expression of iNOS may be functioned through other way but not VEGF way.
