兔脊髓缺血性损害时血糖、内皮素和MDA的变化及异丙酚的保护作用
Changes of plasma glucose,ET-1 and MDA and protective effects of propofol during spinal cord ischemia in rabbits
【摘要】 目的 :观察家兔脊髓缺血性损害时血糖、内皮素、MDA的变化及异丙酚的保护作用。方法 :家兔 1 2只 ,随机分为缺血组和异丙酚组 ,肾下阻断腹主动脉 4 0 min后开放 ,组 II于阻断前静注异丙酚 5mg/ kg继以 2 0 mg·kg- 1· h- 1持续输注至主动脉开放 ,组 I则以同样方法静注等容量生理盐水作对照 ,采不同时相动脉血分别测定血浆葡萄糖 ,内皮素 -1和 SOD、MDA的水平 ,术后观察后肢神经功能和脊髓形态学变化。结果 :异丙酚明显降低脊髓缺血期间的血糖 ,内皮素 -1和 MDA浓度 ,保护 SOD活性 ,改善术后神经功能 ,减轻脊髓的病理学损害。结论 :异丙酚对兔脊髓缺血性损伤有良好保护作用 ,其机制可能与其拮抗缺血期脂质过氧化和高代谢反应有关。
【Abstract】 Objective:To observe changes of plasma glucose ET 1,and MDA and the protective effects of propofol during spinal cord ischemia in rabbits.Methods 12 rabbits were randomly divided into ischemia group and propofol group,The infranenal aortic was removed following clamping for 40 minutes,In group II,propofol was given intravenously 5mg/kg before clamping and continued at a rate of 20mg·kg -1 ·h -1 until unclamping,using normal saline as control in group I.For measurement of level of plasma glucose,ET 1 and MDA,the arterial samples were collected at different interval. After operation,the neurologic outcome were evaluated by blind observers and the morphology of spinal cord were observed.Results:Propofol decreased the level of plasma glucosc,ET 1 and MDA during spinal cord ischemia,and protected markedly activities of SOD,improved neurological outcomes.Conclusion:Propofol could protect the spinal cord from ischemia iniury,the mechanisms are probably related to the inhibition peroxynitrite and decrease of high metablic responses during ischemia.
