酒精代谢产物乙醛对大鼠胚胎中脑细胞增殖分化的影响研究

【摘要】 目的 :为探讨酒精代谢产物乙醛对胚胎发育的作用 ,采用胚胎中脑细胞微团培养研究乙醛对其增殖和分化的影响。方法 :从 13天龄体节数 35 36的鼠胚胎中分离中脑细胞 ,施与不同剂量的乙醛于体外连续培养 5天后 ,测定乙醛对增殖和分化的影响 ,细胞的分化抑制表示有潜在致畸性。结果 :乙醛对中脑细胞增殖和分化的结果 ,显示 0 .1mM及以上剂量即能明显的抑制细胞的增殖分化过程 ,显示出乙醛能阻止神经形成和集落形成 ,其分化抑制剂量 (ID5 0 )和增殖抑制剂量 (IP5 0 )分别为 9.97mM和 6.69mM。结论 :根据的标准乙醛是一种非特异性的细胞增殖分化抑制剂 ,具有潜在的致畸性。

【Abstract】 Objective:To explore the effects of alcohol’s metabolite acetaldehyde on embryonic development,acetaldehyde on proliferation and differentiation were studied by using embryo midbrain cell macromass culture.Method:13-day rat embryo midbrain cells were derived and cultured from brain(CMS)of 35to 36somites of rat embryos .Midbrain cells was exposed to different doses of acetaldehyde.Proliferation and differentiation of neurons from midbrain cells were measured after 5 days of culture in vitro.Inhibition of differentiation was the indicator of potential teratogenicity .Result:The effects of acetaldehyde on differentiation and proliferation of midbrain cells of rat showed that at 0.1mM acetaldehyde and above resuled in inhibition of proliferation and differentiation of midbrain cells significantly.The results showed that acetaldehyde could induce inhibition of neurogenesis and chondrogenesis.ID50 and IP50 of acetaldehyde corresponding were 9.97mM and 6.69mM.Conclusion:Aacetaldehyde is unspecific inhibitor for proliferation and differentiation,according to Renault assessment criteria,acetaldehyde have potential teratogenicitiy.
