鸭乙型肝炎肝纤维化模型的初步研究

【摘要】 目的 :研制乙型肝炎肝纤维化的动物模型。方法 :采用 1d龄樱桃谷鸭 ,随机分为正常组、鸭乙型肝炎病毒 (DHBV )造模组及 CCl4造模组 ,分别于 6 0、10 0及 112 d随机抽样观察两造模组鸭肝组织病理变化 ,于 10 0 d检测 3组动物肝组织 DHBV、DNA、肝功能和肝纤维化指标。结果 :CCl4造模组脂肪变性明显 ,肝细胞水样变性伴中等炎性细胞浸润 ,纤维组织增生明显 ;DHBV造模组以水样变性为主 ,可见肝窦消失或肝窦扩张瘀血 ,脂肪变性以小脂滴为主 ,肝细胞坏死程度较轻 ,但炎性细胞浸润明显 ,且以淋巴细胞多见 ,纤维增生形成时间略长 ,6 0 d尚未见纤维增生 ,80 d时汇管区纤维组织中度增生 ,10 0 d和 112 d时纤维组织明显增生 ,有的形成假小叶 ,其纤维化形成率与 CCl4组 (90 % )接近 (10 0 d为 6 0 .7% ,112 d为 87.5 % )。肝组织 DHBV DNA检测结果 ,DHBV造模组水平较高 ,其肝功能 AL T正常 ,白蛋白明显降低 ,球蛋白显著升高 ,肝纤维化指标如 HA、PC 和肝组织 Hyp均明显上升。结论 :采用 DHBV阳性血清反复攻击樱桃谷鸭 112 d以上 ,可复制形成率较高的鸭乙型肝炎肝纤维化模型

【Abstract】 Objective:To study the animal hepatic fibrosis model with hepatitis B.Methods:49 ducks only one day old were randomly dividedinto three groups:normal group,DHBV model group and CCl 4 group.The pathologic change of ducks of every group was respectively observed using random sampling method in sixty days,one hundred days, one hundred and twelve days.Hepatic fibrosis indexes of those ducks liver tissue,such as DHBV, DNA, liver function were detected in one hundred days.Results:In CCl 4 model group steatosis was obvious,liver cells watery degeneration with middle degree inflammatory cell infiltration and clear fibroplastic proliferation;In DHBV model group liver cell tack watery degeneration as chief change with disappear or stasis of hepatic sinusoid and steatosis mostly showed small fatty drops.Their necrotic degree was light but inflammatory cell infiltration was clear especially lymphocyte infiltration.The forming time of fibroplastic proliferation was little longer than other group. In sixty days fibrous proliferation was not formed, yet in eighty days middle degree proliferation in portal area but in one hundred days and one hundred and twelve days fibroplastic proliferation was distinct,some formed pseudolobuli. The forming rate of fibrosis(100 days 60.7%,112 days 87.5%) was nearly CCl 4 group(90%).DHBV,DNA of hepatic tissue showed:DHBV model group level was higher than those of the CCl 4 model group.Liver function and fibrosis indexes showed:ALP was normal but albumin markedly lowered,globulin markedly rose,at same time,HA,PCⅢ and Hyp of hepatic tissue also significantly rose.Conclusion:The ducks were infected repeatedly using DHBV positive serum,more than 112 days,can replicate hepatic fibrosis model with DHBV.
