三七总皂甙对急性有机磷农药中毒脏器损伤的保护作用研究
Protective effects of panax notoginseng on organ damage of acute organic phosphorus poisoning in rabbits
【摘要】 目的 :研究三七总皂甙对急性有机磷农药中毒脏器损伤的保护作用及其机制。方法 :健康日本大耳兔2 4只 ,随机分为 3组 ,每组 8只 ,正常对照组 (A组 )输入与其它 2组等量 (40 ml)的生理盐水 ;急性有机磷农药中毒组 (B组 )胃管灌入敌敌畏乳剂 16 m g/ kg;三七总皂甙保护组 (C组 )在灌入敌敌畏前 30分钟静脉给予三七总皂甙 10 0 mg/ kg,观察 3组动物血清天冬氨酸转氨酶 (AST)、丙氨酸转氨酶 (AL T)、乳酸脱氢酶 (L DH)、肌酸磷酸激酶 (CPK)、尿素氮 (BU N)、肌酐 (Cr)、胆碱酯酶 (Ch E)、肿瘤坏死因子 (TNF) ,白介素 6 (IL 6 )和 IL 8不同时期的变化值 ,同时观察 8小时兔存活数和心、肝、肾病理学变化。结果 :B组 AST、AL T、L DH、CPK、BU N、TNF、IL 6、IL 8较 A组、C组显著升高 (P<0 .0 5和 P<0 .0 1) ,Ch E较 A组显著减少 (P<0 .0 1) ,与 C组比较变化不大 (P>0 .0 5 )。8小时兔存活数 ,A组 8只 ,B组 2只 ,C组 6只。病理学改变 ,B组心、肝、肾组织损伤程度大于 C组。结论 :三七总皂甙可能通过抑制 TNF和 IL 的释放对急性有机磷农药中毒致脏器损伤起到保护作用。
【Abstract】 Objective:To investigate the protective effects of panax notoginseng on organ damage caused by acute organic phosphorus poisoning.Methods:Twentyfour healthy Japanese white rabbits were randomly divided into 3 groups,which were group A (control),group B (organic phosphorus poison,that was D.D.V.,was given through gastric canal at a dosage of 16 mg/kg) and group C (panax notoginseng was given intravenously 30 minutes prior to the use of D.D.V.at the dosage of 100 mg/kg).Plasma contents of aspartate aminotransferase(AST),alanine aminotransferase(ALT),lactate dehydrogenase(LDH),creatine phosphokinase(CPK),blood urea nitrogen(BUN),creatinine(Cr),cholinesterase(ChE),tumor necrosis factor(TNF),interleukin6(IL6) and IL8 were measured.Pathological changes in heart,liver and kidney were investigated and the survival rate of the rabbits was calculated.Results:①The plasma contents of AST,ALT,LDH,CPK,BUN,TNF,IL6 and IL8 were significantly increased in group B as compared with those in group A and C ( P <0 05 and P <0 01,respectively).The level of ChE in group B was significantly lower than that of group A ( P <0 01) and was not significantly lower than that of group C( P >0 05).②The survival rate of group B and C were 25 0% (2/8) and 75 0% (6/8).③Pathological changes of heart,liver and kidney were more obvious in group B than those of group C.Conclusions:Panax notoginseng has protective effects on the organ damage caused by acute organic phosphorus,the mechanism of which might be related to its inhibitory effects on TNF and ILs secretion.
