眼镜王蛇伤抗蛇毒血清治疗的实验研究

【摘要】 目的:探讨用抗眼镜蛇毒与抗银环蛇毒血清配伍治疗眼镜王蛇伤的治疗方案。方法:改良雷克健氏法分离纯化眼镜王蛇毒;微量免疫电泳分析5 种国产抗蛇毒血清与眼镜王蛇毒组分的共同抗原性;对2 种同科属及1种非同科属的抗蛇毒血清进行同科属及非同科属血清配对动物体内保护实验;用孙氏综合法进行LD50 实验。结果:眼镜蛇毒LD50 为(0.360±0.045)m g/kg。微量免疫电泳证实,抗眼镜蛇毒血清和抗银环蛇毒血清分别与眼镜王蛇毒在Ⅰ、Ⅱ、Ⅵ、Ⅶ、Ⅸ、Ⅹ、ⅩⅠ、ⅩⅡ及Ⅰ、Ⅱ、Ⅵ、Ⅶ组分有沉淀线。用抗眼镜蛇毒与抗银环蛇毒血清配伍治疗眼镜王蛇伤在各用0.1 m l的小剂量下即可达到100% 的保护率,优于单用一种抗蛇毒血清以及与其它抗蛇毒血清配伍。中毒程度越深,所需2 种抗蛇毒血清的量就越大。结论:用抗眼镜蛇毒与抗银环蛇毒血清配伍可协同治疗眼镜王蛇伤,优于用其它抗蛇毒血清配伍治疗眼镜王蛇伤的治疗方案。

【Abstract】 Objective:To investigate the therapeutic effect of combinations of naja naja antivenin and bugarus multicinctus antivenin on ophiophagus hannah bite.Methods:According to KJ Lei′s method,the ophiophagus hannah venom was separated and purified,and microimmunoelectrophoretic test was used to analyze the common antigen character of purified gradients of ophiophagus hannah and five native snake venoms.Protective test in vivo was performed in animals to compare the therapeutic effects of combinations of any two of three antivenins.In addition,SY Shun′s method was used in LD 50 test.Results:The LD 50 of ophiophagus hannah venom was (0 360±0 045)mg/kg.Microimmunoelectrophoretic test showed that naja naja venom had common antigen with gradients Ⅰ,Ⅱ,Ⅵ,Ⅶ,Ⅸ,Ⅹ, ⅩⅠ , ⅩⅡ of ophiophagus hannah venom,and gradients Ⅰ,Ⅱ,Ⅵ,Ⅶ of bugarus multicinctus venom,respectively.A combination of naja naja antivenin and bugarus multicinctus antivenin offered 100% survival rate in the treatment of ophiophagus hannah bite even in a low dose (0 1 ml each).It was more effective than the use of any one type of antivenin alone or other combinations of antivenins.More severe the venenation was,higher dose of antivenin was needed.Conclusions:Combination of naja naja antivenin and bugarus multicinctus antivenin appears to be effective in the treatment of ophiophagus hannah bite.
