用伏安型生物传感器识别微生物细胞的研究
The Study on the Classification of Microbial Cell by a Voltammetric Biosensor
【摘要】 〔目的〕探讨一种利用伏安型生物传感器快速识别微生物细胞的新方法。〔方法〕所用传感器由一个平面热解石墨电极、铂电极及 Ag/ Ag Cl电极的三电极系统组成。将阻留微生物细胞的滤膜紧附在工作电极表面 ,然后在工作电极与对极间施加一扫描电压 ,进行半微分循环伏安扫描 ,记录伏安图谱 ,依据峰电位识别微生物。〔结果〕本文对啤酒酵母菌、大肠杆菌和枯草芽孢杆菌三种微生物进行了识别研究 ,研究发现它们的峰电位各不相同 ,分别是 0 .75、 0 .85和 0 .6 9V (vs Ag/ Ag Cl饱和氯化钾 )。〔结论〕提示可利用峰电位进行微生物细胞的识别 ,方法极为简便快速。
【Abstract】 睴bjective〕 The paper studies the method of the classification of microbial cell by a voltammetric biosensor.〔Method〕 An electrochemical method for detection of microbial cells has been developed by appling semi-differential cyclicde voltammetry to microbial cells.A modified basal plane pyrolytic graphite electrod was employed as a working electrod.Apply a sweep voltage between a working electrode and a counter electrode while the microbial cells on a membrane filter are brought into contact with the working electrode, Then measure the generated potential between the electrodes in phosphate buffered soline.〔Result〕 The anodic peak potential of S.cerevisiae,B.Subtilis and E.Coli are 0.75V,0.69V,0.85V(vs Ag/AgCl)respectively.Therefore,these bacteria can be clearly distinguished from each other.The effects of modifiers was also discribed in this paper.〔Conclusion〕 We will apply this system to discrimination of other microbial cells.
