【摘要】 目的 探讨术前区域动脉灌注化疗对人结直肠癌细胞增殖的影响。方法 选择术前临床病理证实的结直肠癌患者 20例，于术前选择肿瘤供应动脉行区域灌注化疗， 10 d后行根治手术。免疫组织化学法测定并比较癌组织增殖细胞核抗原（ PCNA）及细胞周期蛋白 D1（ cyclin D1）的表达水平。同时选择同期未行动脉灌注化疗的结直肠癌患者直接接受根治手术，作为对照组。结果 化疗组及对照组 PCNA标记指数 (LI)分别为 16. 4％± 15. 2％及 38. 1％± 17. 8％（ P=0.02） ; cyclin D1 LI分别为 8. 7％± 8. 0％ 及 27. 4％± 16. 5％（ P=0. 0002)。细胞增殖指标 PCNA及 cyclin D1的表达在化疗组显著低于对照组。结论 术前选择性区域动脉灌注化疗对结直肠癌细胞增殖可能有较明显的抑制作用，从而发挥抑制肿瘤生长的治疗作用。更多还原

【Abstract】 Objective To investigate the influence of preoperatively intraarterial infusion chemotherapy on cell proliferation of colorectal cancer. Methods Twenty patients with colorectal cancer receiving preoperatively intraarterial infusion chemotherapy were selected as Group A. Another 20 patients with colorectal cancer and without any preoperatively adjuvant therapy were studied as control, Group B. With LSAB method, all of their tumor samples were immunohistochemically stained to determine expression level of proliferating cell nuclear antigen (PCNA) and cyclin D1. Results Expression of PCNA and cyclin D1 were measured by labeling index (LI, positive cells per 1000 tumor cells). Lower expression of PCNA and cyclin D1 were demonstrated in Group A than in Group B (for PCNA 16. 4％± 15. 2％ vs.38. 1％± 17. 8％ , P=0.02 ; and for cyclin D1 8. 7％± 8. 0％ vs 27. 4％± 16. 5％ , P=0. 0002). Conclusions Cell proliferating activity of colorectal carcinoma was suppressed prominently after preoperatively intra- arterial chemotherapy. And it may benefit from high- dose regional chemotherapy. Whether this adjuvant chemotherapy may induce apoptosis, and its potential benefits to survival are awaited for further investigation.更多还原
