实验性大鼠肝癌间质差异形成的机制

【摘要】 目的 用大鼠肝细胞性肝癌 (HCC)和胆管细胞性肝癌 (CC)模型 ,观察肝癌细胞外基质 (ECM)及间质细胞 ,探讨HCC和CC间质差异形成的可能机制。方法 用免疫组织化学方法分别显示I型胶原 (ColⅠ )、Ⅲ型胶原 (ColⅢ )、结蛋白 (DM)和α 平滑肌肌动蛋白 (α SMA)在肝纤维化、HCC和CC的分布。结果 在肝纤维化和肝癌中 ,DM及α SMA阳性细胞与ColⅠ、ColⅢ的分布具有一致性 ,即DM及α SMA阳性细胞的区域可见胶原沉积。CC的DM、α SMA阳性细胞及ColⅠ、ColⅢ多于HCC者。结论 DM及α SMA阳性细胞在肝癌间质产生中具有重要作用。HCC和CC间质ColⅠ、ColⅢ量的差异可能与DM及α SMA阳性细胞的数量和活性有关。上述实验可反映人HCC和CC间质量差别的形成机制

【Abstract】 Objective To observe the extracellular matrix and stromal cells in rats’ hepatocellular carcinoma (HCC) and cholangiocarcinoma (CC) models, and to explore the possible mechanism of stromal difference in the two types of primary liver cancer.Methods By using histochemistry method to study the distribution of collagen I(Col Ⅰ), collagen Ⅲ(Col Ⅲ), desmin(DM) and α smooth muscle actin (α SMA) in hepatic fibrosis, HCC and CC.Results The distribution of DM, α SMA positive cells were consistent with that of Col Ⅰ and Col Ⅲ in hepatic fibrosis and liver cancer, namely, within the DM and α SMA positive cells zone, one could see collagen deposition. The DM, α SMA positive cells and Col Ⅰ, Col Ⅲ were more abundant in CC than in HCC.Conclusion The present experiment shows DM, α SMA positive cells play important role in hepatocarcinomatous stroma, the difference in quantity of Col Ⅰ, Col Ⅲ in HCC and CC might be related to the number and activity of DM and α SMA positive cells. The above experiment can reflect the stromal difference in human HCC and CC.
