三维显示视皮层长时增强过程中DNA/RNA分布的定量分析
Quantitative analysis of DNA/RNA in the visual cortical during the increasing processusing LSCM 3D visualization
【摘要】 激光扫描共聚焦显微镜可对数百 μm厚的组织切片进行共聚焦扫描 ,获得连续的共聚焦平面图像系列 ,通过计算机三维重建算法得到相应组织的立体图像。本文使用 15- 2 0天龄幼年大鼠视皮层脑片的标本 ,在通氧状态下 ,用荧光探针AO染色脑片 ,激光扫描共聚焦显微镜对全脑片不同层面进行共聚焦断层扫描 ,沿纵轴每 2 0 μm扫一次 ,共扫描 16次。再利用总值方式的投影算法对其三维重建 ,得到幼年大鼠视皮层脑片的DNA/RNA分布的立体图像。
【Abstract】 In the experiment ,Using the visual cortical brain slices of Sprague-Dawley (SD)rats aged 15-20 days,the brain slices were dyed with Acridine Orange(AO)in vitro.With the LSCM,the brain slices were scanned into 16 optical sections in an increment of 20 microns along Z-axis,and the three-dimension image of the DNA/RNA of the brain slices was analyzed guantitatively for the changes of DAN/RNA in different stratum and different layers of any slice.Finally,We chtained the 3D image of visual cortical DNA/RNA in young rat brain section.
