【摘要】 采用原位杂交技术结合计算机图像处理技术 ,观察褪黑素 ( MEL)对大鼠脑内某些核团内神经细胞的前强啡肽原 ( PPD) m RNA表达的影响。实验大鼠分 2组 ,给药组大鼠间隔 1 2 h腹腔注射 MEL2次 ,每次剂量 90 mg/ kg,对照组注射配药液。于最后一次注射 1 2 h后 ,灌注取脑、冰冻切片 ,进行原位杂交实验 ,计算机图像处理技术测定染色脑片积分光密度 ( IOD)值。结果观察到 ,给药组大鼠视上核 ( SON)、下丘脑室旁核( PVNH)、中缝背核 ( RD)内神经细胞的 PPD m RNA表达明显强于对照组 ;其 IOD值均显著高于对照组 ( P<0 .0 1 ) ,而在中脑导水管周围灰质腹外侧区未见显著变化 ( P>0 .0 5)。上述结果表明 ,MEL可促进 SON、PVNH、RD内 PPD m RNA表达。

【Abstract】 By using in situ hybridization histochemistry technic and computer assisted image processing and analyzing technique,the present study was designed to observe the altered expression of preprodynorphin(PPD) mRNA to be localized in neurons in some brain nuclei following the exogenous administration of melatonin.The experimental rats were divided into two groups,injected intraperitoneally two times at 12 h interval with melatonin at 90 mg/kg of body weight and with vehicle,respectively.Twelve hours after the last injection,the rat brains were processed for coronal sections and nonradioactive in situ hybridization histochemistry technic was used. The integral optical density(IOD) values of the stained brain sections were measured by using the computer image processing technique.It was observed that the PPD mRNA expression in supraoptic nucleus(SON), paraventricular nucleus of hypothalamus (PVNH) and nucleus raphe dorsalis(RD) was stronger in the brain sections of rats treated with melatonin than in those treated with vehicle.The IOD values in the above nuclei were higher in the melatonin treated group than in the vehicle treated group (P<0 01).In the periaqueductal gray,there was no difference (P>0 05) about the IOD values between the melatonin treated group and the vehicle treated group.These results demonstrate that melatonin may enhance the expression of PPD mRNA in SON,PVNH and RD.
