白斑综合症病毒对斑节对虾亲虾的感染及垂直传播的初步研究

【摘要】 白斑综合症病毒 (WSSV)对斑节对虾 (Penaeusmonodon)亲虾的致病力强 ,其致病性特征与对未成熟个体的致病性特征相同 ;经WSSV感染的斑节对虾亲虾的死亡率为 10 0 % .感染了WSSV的亲虾的PCR检测结果均呈阳性 ,组织切片观察可见到明显的组织病变 ,电镜观察发现病变组织 (包括卵巢 )的细胞内有大量病毒粒子 ;原位杂交结果证明 ,患白斑综合症的斑节对虾亲虾的卵巢中不仅卵母细胞感染上病毒 ,卵细胞也感染上了病毒 .这些结果表明 ,WSSV在斑节对虾中具备垂直传播的条件 .

【Abstract】 The infectivity of white spot syndrome virus(WSSV) to the brooders of black tiger prawn, Penaeus monodon, was studied by means of injection of prawn virus extract The result indicated that WSSV infection was characterized by a wide range of target tissues originating from both the mesoderm and the ectoderm, rapid diseases onset and high mortality in brooder, just like as in immature To identify the vertical transmission route of WSSV, we conducted a combine study using currently available nucleic acid diagnostic tools, including polymerase chain reaction (PCR) and in situ hybridization, and conventional histological observations using light (LM) and transmission electron (TEM) microscopy to examine the sites for virus multiplication. The tissue/organ samples detected included gills, stomach, abdominal muscle, ovary, midgut, hepatopancreas, pleopods, etc The most significant result was that both oocytes and mature egg cells were WSSV positive in the ovaris of brooders infected by WSSV This result represented that the vertical transmission of WSSV might exist in Penaeus monodon
