大鼠心肌IP第二窗对I/R心肌细胞凋亡的影响及对bcl-2、bcl-xl表达的调节
Effect of ischemic preconditioning on cardiac myocyte apoptosis and the regulation of IP on Bcl-2 and Bcl-xl during myocardial ischemia / reperfusion course in rat
【摘要】 目的 :研究缺血预适应 (IP)第二保护窗对大鼠缺血再灌注 (I/R)心肌细胞凋亡的影响及其对凋亡抑制基因bcl- 2和bcl-xfl蛋白表达的调节。方法 :采用TUNEL法和免疫组化方法。结果 :(1)IP +IR2 4h组TUNEL法阳性心肌细胞核数量及阳性心肌细胞核占总心肌细胞核数的百分比均明显少于IR2 4h组 (P <0 .0 5 - 0 .0 1) ;(2 )IP +IR2 4h组表达bcl- 2和bcl-xl蛋白阳性的心肌细胞数及阳性心肌细胞占心肌细胞总数的百分比均明显高于IR2 4h组 (P <0 .0 5 - 0 .0 1)。结论 :(1)大鼠心肌IP第二保护窗能够显著减少I/R心肌细胞凋亡 ;(2 )IP通过上调凋亡抑制基因bcl- 2和bcl-xl蛋白表达可能是其减少I/R心肌细胞凋亡的机制之一
【Abstract】 Objective:To explores the effect of ischemic preconditioning on cardiac myocyte apoptosis and the regulation of which on bcl-2 and bcl-xl during myocardial ischemia/reperfusion course in rat. Methods:TUNEL and immunohistochemical method. Results:(1) The numbers of positive cardiac myocyte nuclear and the percentage of positive cardiac myocyte nuclear in IP+IR24h group decreased significantly(P<0.05) compared with that of IR24h group;(2) The numbers of protein bcl-2 and bcl-xl positive cardiomyocyte and the percentage of protein bcl-2 and bcl-xl positive cardiomyocyte in IP+IR24h group were higher(P<0.05) compared with those of IR24h group,respectively. Conclusion:(1) The second window of IPs protection could decrease cardiomyocyte apoptosis significantly and (2) Up-regulating the protein expression of bcl-2 and bcl-xl in cardiomyocytes during I/R course may be one of the mechanisms of second window of IPs protection.
