淫羊藿甙逆转转化生长因子β2对LAK、CD3AK细胞的免疫抑制作用
Reversal of transforming growth factor β2 mediated immunosuppression of LAK and CD3AK by Icarrin
【摘要】 探讨淫羊藿甙逆转转化生长因子 β2 (TransformingGrowthFactorβ2 ,TGFβ2 )对LAK、CD3AK细胞的免疫抑制作用及其机制。方法 :采用MTT法 ,RT PCR和流式细胞术。结果 :淫羊藿甙能逆转受TGFβ2 抑制的LAK和CD3AK细胞的杀伤活性 ,并能部分恢复受TGFβ2 抑制的LAK细胞表面IL 2Rα表达和CD3AK细胞内穿孔素mRNA水平 ,以及CD3AK细胞的增殖活性。结论 :淫羊藿甙通过恢复LAK细胞IL 2Rα表达 ,提高CD3AK细胞内穿孔素mRNA水平及促进CD3AK增殖 ,逆转TGFβ2 对LAK、CD3AK的免疫抑制作用。
【Abstract】 Objective:To study the reversal of transforming growth factor β 2(TGFβ 2) mediated immunosuppression of LAK and CD 3AK cells by Icarrin and its mechanisms.Methods:MTT assay, RT-PCR, Flow Cytometry.Results:ICA can reverse TGFβ 2 mediated suppression of the cytotoxic activity of LAK and CD 3AK cells.It can also reverse TGFβ 2 mediated downregulation of IL-2Rα expression on LAK cells and perforin mRNA level in CD 3AK cells.Conclusion:The mechanisms of ICA function is probably due to its capability of reversal TGFβ 2- mediated suppression of IL-2Rα expression on LAK cells and perforin mRNA level in CD 3AK cells.
