IL-12对狼疮肾炎PBMC中信号传递分子STAT3、STAT4的作用
Effects of IL-12 on signal transduction molecules STAT3 and STAT4 in PBMC in lupus nephritis
【摘要】 目的 :为了探讨IL 12对狼疮肾炎PBMC发挥生物学作用的细胞内信号传递机制。方法 :采用MTT、免疫沉淀、免疫印迹等方法 ,检测IL 12对PBMC增殖及STAT3、STAT4的作用 ,并比较在狼疮肾炎活动期、静止期的差别。结果 :发现与正常人对照及静止期相比 ,IL 12对活动期PBMC不需要IL 2的预诱导 ,可促进PBMC增殖并在短时间内激活STAT3、STAT4,而IL 2对STAT4无作用。结论 :表明IL 12对PBMC的作用可通过STAT3、STAT4途径发挥 ,活动期PBMC存在异常活化 ,这可能是IL 12促进狼疮肾炎PBMC参与发病的细胞内机制之一
【Abstract】 Objective: To investigate the mechanism of signal transduction of IL 12 on peripheral blood mononuclear cell (PBMC) in lupus nephritis (LN).Methods: The authors measured proliferation of PBMC by MTT. STAT 3 and STAT 4 were detected by immunoprecipation and Western Blot Technique. Results:In contrast to normal group and the static stage of LN, IL 12 alone stimulated DNA synthesis and activated STAT 3 and STAT 4 in absence of IL 2 in short time in PBMC in the active stage of LN, wherease IL 2 had no effect on STAT 4 but STAT 3.Conclusion:The effect of IL 12 in PBMC may be mediated through the activation of STAT 3 and STAT 4, PBMC may have been abberantly activated in the active stage of LN, which may be a mechanism of IL-12 to take part in the pathogenesis of LN.
