实验性全脑缺血再灌流CNS bcl-2基因变化研究
Expression of Bcl-2 Gene in Brain after Repetitive Ischemia-Refusion
【摘要】 目的 探讨bcl 2基因表达在全脑缺血再灌流不同时间的变化规律 .方法 本试验在对小鼠双侧颈总动脉阻断再灌流后 1,3,7,14,2 8,42d分别取额叶皮层海马区脑组织 ,采用免疫组织化学染色 ,逆转录PCR技术对bcl 2基因表达进行检验 .结果 bcl 2基因在正常神经细胞中有较弱表达 ,再灌流 1d后其表达开始增加 ,至 7d时达高峰并持续至 14d ,与对照组有显著差异 (P <0 .0 1) .结论 bcl 2基因抑制细胞凋亡的发生 ,其表达水平与细胞凋亡存在密切关系 .
【Abstract】 Objective To investigate the expression of bcl 2 gene in brain of rats after repetitive ischemia refusion.Methods Bcl 2 genetic expression of frontal cortex and hippocampus of rats is taken on 1st day,3rd day,7th day,1st week,2nd week,4th week,6th week respectively after repetitive ischemia refusion by means of immunohistochemical technique and RT PCR technique.Results Expression of bcl 2 gene in normal neurocyte increased expression appear after ′st day to top on 7th day and keep it at highest level until two weeks,and it is significant differance from control group ( P <0.01).Conclusion There is close relation between protein expression level of bcl 2 gene and neurocyte apoptosis.
