高效液相色谱法测定大豆提取物中大豆异黄酮的含量
Determination of Isoflavone in Extract of Soybean by HPLC
【摘要】 本文建立了大豆提取物中大豆异黄酮的高效液相色谱测定方法 ,通过正交试验确定了样品水解最佳条件为 :1 .0mol/lHCl-MeOH溶解、80℃下、回流水解 0 .5h ,采用Nova -PakC183.9× 1 50mm 4 μm色谱柱 ,MeOH -0 .4 %H3PO4( 4 7:53V/V)为流动相 ,流速 0 .7ml/min ,检测波长 2 60nm等色谱条件下测定甙元含量 ,通过换算因子计算大豆异黄酮的含量。该方法快速、灵敏、重现性好 ,回收率达 99% ,变异系数小于 3%。为产品的进一步开发研究打下了良好基础。
【Abstract】 A method for determination of isoflavone in extract of soybean by highperformance liquid chromatography(HPLC)is described.The optimum hydrolysis condition of extract of soybean was obtained by using the orthogonal design method and the analysis of variance,the analytical condition of HPLC was also established.The method is rapid,sensitive,reproduciable and provides a recovery of more than 99% with RSD less than 3%.
