【摘要】 目的　探讨药物标记抗体导向治疗老年人免疫性血小板减少性紫癜的可行性及效果。方法　分别用间接和直接交联法将注射用免疫球蛋白 (IVIG)与氨甲喋呤 (MTX)制备成标记抗体 ,用间接免疫荧光法检测标记抗体 Fc段结合活性 ,以 Fc受体阳性的小鼠巨噬细胞和人单核细胞白血病细胞株 U937为靶细胞 ,用 MTT法测定标记抗体的杀伤活性。结果　标记抗体对巨噬细胞的杀伤作用明显大于游离 MTX:标记抗体对 Fc受体阳性细胞的杀伤作用明显大于 Fc受体阴性细胞。间接标记抗体 IVIG- HSV- MTX的特异杀伤作用明显大于直接标记抗体 IVIG- MTX。结论　标记抗体在体外对单核、巨噬细胞显示了相对特异的杀伤活性。更多还原

【Abstract】 To investigate the feasibility and effect of guide treatment of drug lablled antibody on immune thrombocytopenic purpura.Methods IVIG and MTX were made into lablled antibody by direct and indirect conjugating methode respectivity.Binding activity of lablled antibody Fc part was detected by the indirect immune fluorescense.Phagocyte of mouse and U937-the leukemia cell line of human monocyte whose Fc receptor is positive were used as target cells.The killer activity of lablled antibody was tested by MTT methods.Results Lablled antibody showed stronger killer effect upon phagocyte than free MTX.Lablled antibody show stronger killer effect on Fc receptor positive cells compared with the negative one.The specific killer effect of indirect lablled antibody IVIG IIAS MTX was significantly stronger than that of direct lablled antibody IVIG MTX.Conclusion Lablled antibody showed relatively specific killer effects on monocyte phagocytes in vitro更多还原
