薏苡仁酯对人鼻咽癌细胞辐射效应的协同作用
The Enhancement of Coixenolide in Irradiation to Human Nasopharyngeal Carcinoma Cell
【摘要】 目的 探讨薏苡仁酯 (coixenolide ,CXL)对人鼻咽癌细胞CNE 2Z辐射效应的影响。方法 以60 Co为放射源 ,采用微量细胞克隆形成法检测CNE 2Z对γ射线的敏感性。结果 CXL使CNE 2Z的辐射—存活曲线向左移 ,Do、Dq 和N值下降。不同浓度的CXL (5 2 0 μmol L)使放射剂量减少 17 2 6 %— 4 1 90 % (在D3 7水平 ) ,其增效比 (ER ) :Do 比值 1 14 1 6 4 ,Dq 比值1 2 7— 1 79。另外 ,CXL和射线的合并效应大于两者单独效应之和。结论 CXL和射线的相互作用产生协同效应
【Abstract】 Objective To study the effect of coixenolide (CXL) on irradiation to human nasopharyngeal carcinoma (NPC) cell line CNE 2Z. Methods microcolony formation assay was used in determining susceptibility of CNE 2Z to γ ray.Results CXL shifted the radiation dose response curve to the left,with the decrease of D o、D q and N values.Radiation doses reduced by 17 26%~41 90% at the D 37 level,the enhancement ratios were 1 14~1 64 and 1 27~1 79 at D o and D q level respectively,when different doses (5~20μmol/L) of CXL were added to culture system.In other experiments,2Gy of ray and 10μmol/L of CXL produced 5 21% and 8 02% of inhibition to colony formation by CNE 2Z respectively,and then,combination of CXL and irradiation with the same dose showed 22 53% of suppression (not 5 21%+8 02%=13 22%).Conclusion Combining CXL and γ ray causes synergism in depressing the proliferation of CNE 2Z.
