神经胶质生长因子促进受损周围神经功能恢复
Glial Growth Factor Accelerates Functional Recovery of Injured Peripheral Nerve
【摘要】 目的 评价重组体人类神经胶质生长因子 2 (rhGGF2 )对大鼠坐骨神经挤压伤后运动功能恢复的效果。方法 :73只大鼠分 3组。第 1组 (n =5 )实施模拟挤压操作。第 2组 (n =34)、第 3组 (n =34) ,每只动物选一 5mm长的坐骨神经段承受 10 0 g挤压 2h。第 3组为治疗组 ,rhGGF2 1mg/kg挤压后皮下注射 ,每日 1次 ,连续 4d。第 2组用等量的载体治疗。通过计算坐骨神经功能指数 (SFI)和测量趾长伸肌强直收缩力来评价神经运动功能恢复。结果 :第 3组神经在术后 11~ 2 1d表现出显著的SFI改善。第 3组肌肉收缩力强于第 2组 ,从 4~ 14d在 70Hz ,10 0Hz刺激下有显著性差异。组织学切片显示 3组呈现较轻的轴突溃变和较早的神经再生。结论 :rhGGF2治疗坐骨神经挤压伤能有效地促进神经再生 ,进而显著改善其功能的恢复
【Abstract】 Objective:To evaluate the effect of recombinant human glail growth factor Ⅱ(rhGGF2) on recovery of motor function of rat sciatic nerve following crush injury.Methods:73 rats were divided into three groups.A sham operation was performed in group Ⅰ(n=5).A 5-mm segment of sciatic nerve was subjected to a 100g crush load for 2 hours in groups Ⅱ (n=34) and Ⅲ (n=34).Group Ⅲ was treated with rhGGF2 (1mg/kg by subcutaneous injection after crush for four days),and group Ⅱ was given the same volume of vehicle,Motor functional recovery was assessed by calculating the sciatic functional index(SFI) and measuring tetanic contractile force of extensor digitorum longus (EDL).Results:Group Ⅲ exhibited a significant improvement of SFI on postoperative day 11 to 21 when compared to group Ⅱ.The EDL contractile force was generally stronger in group Ⅲ than in group Ⅱ,with a significant difference at 70 and 100 Hz stimulus frequencies from day 4 to 14.Histological sections of nerve revealed less axonal degeneration and earlier regeneration of nerve in group Ⅲ.Conclusion:Treatment with rhGGF2 is effective in promoting nerve regeneration,as a result,can significantly improve the functional recovery of rat sciatic nerve following crush injury.
