急性脑梗死患者血小板内钙和TXB2含量与聚集关系的探讨
Relationships cytosolic calcium levels, thromboxane B2 levels of intraplatelet and platelet aggrepation induced by ADP in acute cerebral infarction
【摘要】 目的探讨急性脑梗死急性期患者血小板内游离钙和TXB2含量之间的相互关系及意义。方法 用Fura-2/AM荧光技术测定了46例急性脑梗死患者急性期及25例正常对照组血小板内钙含量,用放射免疫分析法测定其血小板内血栓素B2(TXB2)含量,用比浊法测定其血小板聚集。结果患者血小板内游离Ca2+和TXB2含量均分别明显高于正常对照组(147.26±10.85 vs 119.30±10.94 nmol/L和3.38±0.78vs 4.88±0.97ng/109血小板,P均<0.01);在患者中,低(0.5μmol/L)和高(5μmol/L)浓度二磷酸腺苷(ADP)诱导的血小板聚集均明显高于正常对照组(0.44±0.19vs 0.26±0.14和0.73±0.13vs0.39±0.19,P均<0.01);在患者中血小板内游离Ca2+含量和TXB2含量均分别与5μmol/L二磷酸腺苷诱导的血小板呈明显的正相关性(r分别为0.786和0.593,P均<0.01),而血小板内Ca2+与TXB2成明显的正相关性(r为0.652,P<0.01)。结论 急性脑梗死患者在急性期血小板功能处于活跃状态,可能与其内 Ca2+含量增加、TXB
【Abstract】 Objective To investigate the relationships and the significantly among cytosolic calcium levels thromboxane B2 levels of intraplatelet and platelet aggregation induced by ADP in the patients with acute cerebral infarction (ACI). Methods Cytosolic calcium levels were measured in Fura - 2/AM loaded platelet in 46 patients with ACI and in 25 normal controls, platelet thromboxane B2 levels were investigated by radioimmunoassy (RIA)and platelet aggrega- tion induced by ADP were measured with lumi - aggregometer M450. Results The intraplatelet levels of calcium and thromboxane B2 in the patients with ACI were significantly higher than those in the nounal controls, respectively (147.26±10.85 vs 119.30±10.94 nmol/L, and 3.38±0.78 vs 4.88±0.97 ng/109 platelet, respectively, all P < 0.01 ), but platelet induced by 0. 5 μmol/L ADP and 5 μmol/L ADP in the patients were significantly lower than those in normal con- trols, respetively(0.44±0. 19 vs 0.26 ±0. 14, and 0.73± 0. 13 vs 0. 39 ± 0. 19, respetively, all P < 0.01 ). In the patients, moreover, the intraplatelet calcium levels and intraplatelet thromboxane B2 levels were positively correlated with the aplatelet aggregation induced by 5μmol/LADP (r were 0.786 and 0. 593, respec- tively, all P<0.01 ). And there was a positive relationship between intraplatelet calcium levels and intraplatelet thromboxane B2(r was 0.652 P < 0.01 ).Con- clusions Platelet was activative during the acute phase in the patients with ACI. which was possible related to the increase of cytosolic calcium levels and throm- boxane B2 levels in intraplatelet. Their interaction activated platelet in ACI .These data suggested that using antiplatelet durgs might play an important role in the treatment of ACI.
