PVA固定化细菌ZG-4株生产L-苹果酸的研究

【摘要】 从 60余份土样中筛选得到一株富马酸酶活力较高的细菌ZG 4株。以改性PVA为载体 ,制得固定化细胞。其较佳转化条件 :基质为 1mol /LNa2 Fu,转化液pH 7.0 ,转化温度 40℃。固定化细胞经质量浓度为 0 .4%的胆酸或 0 .0 6%的TPC预处理 ,可有效抑制琥铂酸副产物的生成。添加质量浓度为 0 .1 %没食子单宁 ,可使富马酸酶比活力达 1 .95mmol·h- 1 g- 1 。同时比较了固定化细胞与游离细胞的富马酸酶特性。

【Abstract】 A bacterial strain ZG-4 was screened out from more than 60 soil samples collected from different regions. The whole cells of strain ZG-4 containing high fumarase activity were immobilized in modified polyvinylalcohol (PVA) gel. The optimal pH and temperature for bioconversion of 1M sodium fumarate was 7.0 and 40 ℃,respectively. The formation of succinic acid, by-product during the bioconversion, could be suppressed through pretreatment of biomass by using 0.4% cholic acid or \{0.06%\} TPC. When 0.1% gallotannin was added, the specific activity of fumarase was 1.95 mmol·h\+\{-1\}g\+\{-1\}. Specific property of fumarase for immobilized cells was compared with that of free cells.
