康宁克通对增生性瘢痕Ⅰ、Ⅲ型胶原蛋白及前胶原基因原位表达的影响

【摘要】 目的 探讨康宁克通在活体中对增生性瘢痕Ⅰ、Ⅲ型胶原合成及降解影响的机理。方法 用免疫组织化学及分子生物学技术 ,对 6例增生性瘢痕局部注射康宁克通 3及 7d后 ,Ⅰ、Ⅲ型胶原蛋白及相应前胶原mRNA的原位表达进行了研究。结果 ①注射 7d后 ,Ⅰ型胶原蛋白量降低 (P <0 0 5 ) ,Ⅲ型胶原蛋白量未见明显降低 (P >0 0 5 )。而Ⅰ、Ⅲ型前胶原mRNA在注射后 3d已被明显抑制 (P <0 0 1) ,至注射后 7d ,表达强度进一步下降。②康宁克通局部注射后对Ⅰ型前胶原mRNA的表达抑制可能比对Ⅲ型前胶原强。③Ⅰ、Ⅲ型前胶原mRNA表达在增生性瘢痕中比正常皮肤强。结论 康宁克通对Ⅰ型胶原的抑制比Ⅲ型胶原强。Ⅰ、Ⅲ型胶原mRNA表达在增生性瘢痕比正常皮肤强

【Abstract】 Objective This is to investigate the mechanism of effects of Kenalog on type Ⅰ and Ⅲ collagen syntheses and degradation in hypertrophic scars. Methods Intralesional injection of Kenalog was performed on 6 patients with hypertrophic scars.On the 3rd day and 7th day after the treatment,6 samples from the 6 patients each were collected and type Ⅰ and Ⅲ collagen proteins and in situ procollagen mRNA expressions were studied by means of immunohistochemistry and molecular biology. Results On the 7th day after intralesional injection of Kenalog,type Ⅰ collagen proteins reduced ( P <0.05) and type Ⅲ collagen proteins did not reduced significantly ( P >0.05).On the 3rd day after intralesional injection of Kenalog,both type Ⅰ and type Ⅲ procollagen mRNA expressions were significantly inhibited and on the 7th day both procollagen mRNA expressions decreased further. Conclusions Intralesional Kenalog may inhibit type Ⅰ procollagen mRNA expression more than type Ⅲ. Gene expressive intensions of type Ⅰ and Ⅲ procollagen were higher in hypertrophic scars than in normal skin.
