肿瘤坏死因子α对瘢痕成纤维细胞生物学作用的实验研究

【摘要】 目的 探讨肿瘤坏死因子α(TNF α)对瘢痕成纤维细胞的生物学作用及其在异常瘢痕的发生和防治中的意义。方法 采用组织培养技术 ,建立了成纤维细胞系 ,以MTT法测定细胞活力 ,流式细胞仪检测纤维粘连蛋白 (FN)表达及间接免疫荧光法等技术和方法。对来源于增生性瘢痕和正常皮肤的成纤维细胞进行了对比研究。结果 ①高浓度的TNF α( 10 0 0U/ml ↑ )对增生性瘢痕成纤维细胞活力具有明显抑制作用 ,并使其FN表达减少 11 7% ;②TNF α对正常皮肤成纤维细胞活力具有明显促进作用 ,并使其FN表达增加 5 8%。结论 TNF α对于增生性瘢痕和正常皮肤的成纤维细胞具有不同的生物学作用。研究TNF对增生性瘢痕的调控机制 ,有可能为防治瘢痕的异常增生提供新的方法和途径。

【Abstract】 Objective To investigate the biologic effects of tumour necrosis factor α(TNF α) on scar\|derived fibroblasts and to evaluate the role of TNF α in genesis ,prevention and treatment of scar. Methods Cell strains were established by tissue culture technique. To study the fibroblasts derived from hypertrophic scar and normal dermis,cytoactivity and expression of fibronectin (FN) were determined by MTT,FCM and immunofluorescence. Results① When TNF α is at a high concentration (1?000U/ml ↑),the cytoactivity of hypertrophic scar fibroblasts was decreased significantly,and the positive rate of FN expression was decreased by 11.7%. ② Cytoactivity of normal dermis fibroblasts was increased with TNF α while the positive rate of FN expression was increased by 9.6%. Conclusions TNF α has different biological effects on fibroblasts derived from hypertrophic scar or normal dermis. Studying the regulative mechanisms of TNF on abnormal scar will probably provide a new way to prevent and cure it.
