辅酶Q10对供心热缺血的保护作用

【摘要】 目的 探讨辅酶Q10 对供心热缺血的保护作用 ,以达到延长供心耐受热缺血时限目的。方法 新西兰大白兔 16只随机分为 2组 ,每组 8只。A组为对照组 ,B组为实验组。B组取心前 2周用辅酶Q10 2mg·kg-1·d-1灌胃。热缺血 10min后 ,A组用UW液、B组用UW液加辅酶Q10 液灌洗 ;取心后 ,A组在UW液、B组在UW加辅酶Q10 液中保存 4h。然后在Langendorff模型上 ,A组用氧合KHB液、B组用氧合KHB加辅酶Q10 液再灌注 30min。以功能、代谢和结构等指标评价供心恢复情况。结果 B组各项指标均优于A组。结论 用辅酶Q10 对供心作预处理 ,并在灌洗液、保存液及再灌液中加入辅酶Q10 ,对供心热缺血和冷保存均有良好保护作用 ,值得临床应用。

【Abstract】 Objective This study is to investigate the evidence of protective effect of coenzyme Q 10 (co Q 10 ) on warm ischemia of donor heart. Methods New Zealand rabbits (n=16) were randomly divided into two groups, control group as group 1,experimental group as group 2. The rabbits of group 2 were fed co Q 10 at a dose of 2mg·kg -1 ·d -1 for two weeks before harvesting the heart. After warm ischemia for 10 minutes, perfused with UW solution at 4℃ in group 1 and with UW+ co Q 10 solution at 4℃ in group 2, the hearts were isolated. After that, preservation with UW solution for four hours was given in group 1; while with UW+co Q 10 solution for four hours in group 2. Furthermore, On Langendorff model, Hearts in group 1 were reperfused with KHB fluid while hearts in group 2 with KHB+co Q 10 fluid. "The recovery was evaluated by heart function, cell metabolism and ultrastructure change. Results All indexes in group 2 were obviously superior to group 1. Conclusion Using co Q 10 as pretreatment and as an adjunct in perfusing, preservative and reperfusing solutions has an obvious protective effect on warm ischemia and cold preservation of donor heart.
