唾液中分离的人类疱疹病毒7型的电镜观察及分子生物学鉴定

【摘要】 目的 旨在分离人类疱疹病毒 7型 (HHV 7)的南京地方株。方法 收集健康成人及肾病患儿唾液 ,经抗生素处理和离心过滤后接种于PHA预刺激的人脐带血单个核细胞 (CBMC) ,当细胞出现气球样病变效应 (CPE)时 ,透射电镜观察病毒颗粒、特异性引物扩增病毒核酸并对分离株的PCR扩增产物作序列测定。结果 从 7例唾液标本中分离出 4株病毒 ,电镜下呈典型疱疹病毒科病毒形态 ,经巢式PCR扩增都在预期的 186bp(外引物 )和 92bp(代表株 ,内引物 )处出现条带 ,代表株186bp产物序列与标准株RK和JI株基因相应区域同源性为 10 0 %。结论 分离的病毒为HHV 7。

【Abstract】 Objective The present research intends to isolate Nanjing local strains of human herpesvirus 7 (HHV 7). Methods Saliva were sampled from health individuals and from children with kidney disease. After treatment with antibiotics and filtering, the samples were inoculated to the umbilical cord blood mononuclear cells (CBMCs) stimulated by phytohemagglutinin. When the infected cells presented the typical ballooning and polykaryotic cytopathic effects (CPE), transmission electron microscopy of cells was performed. DNA was extracted and polymerase chain reaction was carried out using the primers specific for HHV 7. The PCR products were also sequenced. Results Four strains were isolated from the seven saliva specimens. Electronic microscopy showed the presence of virus like particles with the characteristic features of herpesviruses. The expected PCR product was amplified from both isolates. The specificity of the reaction was confirmed by DNA sequence and 186bp fragments revealed an identity with HHV 7 RK and JI of 100%. Conclusion The isolated virus was HHV 7.
