阴道加德纳菌ITS-23S rRNA部分基因克隆及序列分析

【摘要】 目的 分析深圳地区阴道加德纳菌 (G .vag)ITS(internaltranscribedspacer) 2 3SrRNA部分基因序列。方法 设计特异性引物 ,建立检测G .vag的PCR方法 ,对临床病人阴道分泌物标本进行检测 ,并对分离出的 4株PCR产物进行分子克隆和序列分析。结果 分离出的 4株G .vagITS 2 3SrRNA基因的核苷酸同源性在 99 5 %以上 ,与Belkum报道株同源性在 98%以上。结论 深圳地区G .vag与国外报道的菌株在ITS 2 3SrRNA基因区变异不大 ,可能属同一基因型。

【Abstract】 Objective To analysis the sequence of ITS 23S rRNA genome of Gardnerella vaginalis from patients with bacterial vaginosis in Shenzhen city. Methods A pair of primers from ITS 23S rRNA genome were designed and used to detect G.vaginalis by polymerase chain reaction method. The partial G.vaginalis genome isolated from 4 patients with bacterial vaginosis were cloned to T vector and sequenced by dideoxy chain termination methods. Results The sequence analysis showed homogeneity of over 99.5% among 4 isolates from Shenzhen and over 98% between Shenzhen isolates and the isolates in Belkums’ report. Conclusion There is no significant difference in ITS 23S rRNA genome of G.vaginalis between the isolates from Shenzhen and those from Belkums’ report.
