八肽胆囊收缩素对神经细胞缺氧的作用

【摘要】 目的 探讨八肽胆囊收缩素 (CCK- 8)对缺氧神经细胞的作用。方法 采用神经细胞原代分离培养技术 ,以细胞外上清液 L DH、神经细胞 3H-谷氨酸摄取及 3H- GABA摄取功能为损伤指标。 结果 缺氧后神经细胞上清液中 L DH含量明显增高 ,3H-谷氨酸和 3H- GABA摄取率较未缺氧对照组明显降低。给予 CCK- 8后 ,可减轻缺氧引起的这些神经细胞损伤 ,CCK- B受体拮抗剂 L- 36 5 2 6 0可阻断 CCK- 8对神经细胞缺氧的保护作用。 结论  CCK- 8对缺氧性神经细胞损伤具有保护作用 ;该作用可被 CCK- B受体拮抗剂阻断 ,说明其保护机理是由 B受体介导产生的。

【Abstract】 Objective To investigate the effects of cholecystokinin octapeptide (CCK 8) on hypoxic damage of neuronal cells. Methods LDH levels in culture medium were measured with LDH kit. The uptake ability of the neuronal cells for the 3H Glutamate and the 3H GABA were used as the markers of the cells damage. Results (1)The LDH concentration in the medium of hypoxic group was increased [from (13.26±2.5 )μ/l to (42.23±4.5) μ/l] significantly attenuated by CCK 8 pretreatment [from (42.23±4.5) μ/l to (20.34±4.23) μ/l], CCK B receptor antagonist L 365260 reversed the CCK 8 effect.(2) 3H Glutamate and 3H GABA uptake rate in the hypoxic group were significantly lower than that in the non hypoxic group. It was significantly increased in the CCK 8 pretreated group, which can be blocked by the CCK B receptor antagonist L 365260. Conclusion CCK 8 can significantly attenuate the neuronal cells damage induced by hypoxia, which may be regulated by its CCK B receptor.
