高糖对大鼠腹膜间皮细胞中p21WAF1基因表达的影响
High glucose stimulates the mRNA expression of p21WAF1 in rat mesothlial cells
【摘要】 目的研究细胞周期调控蛋白p21WAF1在高糖对腹膜间皮细胞增殖抑制中的作用。方法用流式细胞仪技术及半定量逆转录-聚合酶链反应(RT-PCR)方法测定不同浓度的葡萄糖对体外培养的大鼠腹膜间皮细胞细胞周期分布和p21WAF1mRNA表达的影响。结果在高糖的培养条件下,大鼠腹膜间皮细胞出现细胞肥大,胞核增多,细胞分裂停止,且大多数间皮细胞的细胞周期停滞于G1期。在静止期的间皮细胞几乎无p21WAF1mRNA的表达,在高浓度葡萄糖刺激下p21WAF1mRNA明显增加,且随着浓度增高而表达增高,且含糖浓度为3.86%时p21WAF1的表达比含糖为1.38%时明显增高(P<0.05)。在和含糖浓度为3.86%、1.38%组渗透压根同的甘露醇组也几乎无p21WAF1mRNA的表达。结论pp21WAF1可能在高糖对间皮细胞的抑制作用及G1期阻滞中起重要的调节作用。
【Abstract】 Objective To investigate the effect of cell cycle regulator p21WAF1 on the mechanism by which high concentrated glucose inhibits mesothelial cells proliferation. Metkods Flow cytometer technique and semi-quantity RT-PCR method were used to detect the cell cycle distribution and p21WAF1 mRNAexpression of rat mesothelial cells in serum-free DMEM with different glucose concentraion media. Results In the high glucose medium most of the cells became hyperplasia, and had multiple nuclei and arrest of cell division. And most arrested in G1 phase of the cell cycle. The p21WAF1 mRNA was absent in serum-free normal medium. But high glucose stimulated p21WAF1 mRNA expression in dose-dependent way, and the 3. 86% poup was significantly higher than 1. 38% group(P < 0. 05). p21WAF1 mRNA expression was also absent in the D-mannitol groups which had the same osmolarity as the 3. 86% or 1. 38% glucose groups. Conclusion p21WAF1 may be pivotal in the suppression and arrest in the G1 phase of mesothelial cells by the high concentrated glucose.
