早期应答基因产物表达对移植静脉平滑肌细胞增殖的影响

【摘要】 目的 探讨c fos、c jun、c myc等早期应答基因与移植静脉平滑肌细胞 (SMC)增殖的影响。方法 建立Wistar大鼠 (6 0只 )自体血管移植模型 ,移植后 2h、6h、2d、1周、2周、4周分别切取移植静脉 ,应用免疫组织化学方法动态观察早期应答基因的变化情况。结果 正常血管平滑肌细胞中鲜有早期应答基因表达 ,c fos、c jun于移植后 2h达高峰 ,吻合口处为 (13.1± 3.3) %、(12 .8±2 .9) % ,中段静脉为 (10 .3± 2 .8) %、(10 .0± 1.9) % ,与术后 1周相比差异有显著性 (P <0 .0 1) ;c myc于 2h有少量表达 ,于 1周时达高峰 [吻合口 (16 .2± 3.8) %、中段静脉 (11.2± 2 .3) % ],与术后 4周相比差异有显著性 (P <0 .0 1)。结论 在移植静脉中 ,早期应答基因是SMC增殖的早期反应基因 ,参与DNA复制 ;c myc可能参与了细胞的持续增殖。

【Abstract】 Objective\ To investigate the effect of expression of immediate early gene (c fos, c jun, c myc )products on smooth muscle cell (SMC) proliferation in vein graft. Methods\ Autogenous transforming vein models for 60 Wistar rats was developed. The vein grafts were harvested at 2 h, 6 h, 2 days, 1 week, 2 weeks and 4 weeks after grafting. The change of protein of immediate early gene was detected by using immunohistochemistry. Results\ There was little expression in normal vessels. The protein of c for and c jun peaked at 2 h after the grafting, (13.1±3.3)%, (12.8±2.9)% in anastomasis and (10.3±2.8)%, (10.0±1.9)% in middle of the vein graft showing a significant difference between 2 h and 1 week ( P <0.01). The protein of c myc peaked at 1 week (16.2±3.8)% in anastomasis and (11.2±2.3)% in middle of the vein graft and there was significant difference between 1 week and 4 week ( P <0.01). Conclusion\ In vein graft, immediate early gene is an early responsive gene in SMC entrance from silent to proliferation stage. Immediate early gene perhaps participates in replication of DNA.
