胶质瘤多药耐药细胞系C6/adr的建立及其耐药性逆转的研究

【摘要】 目的 研究胶质瘤多药耐药 (MDR)的发生机理及逆转方法。方法 建立了C6 /adrMDR细胞系 ,经RT PCR及免疫组化染色分别研究了mdr l基因及P 糖蛋白 (P gp)的表达 ;采用MTT药敏试验及HPLCA测定细胞内阿霉素浓度的方法研究了异搏定、红霉素、潘生丁、P gp单克隆抗体、复方丹参对MDR的逆转作用。结果 C6 /adrMDR细胞系mdr l基因阳性 ,P gp高度表达。异搏定 (2~ 6 μg/ml)、红霉素 (5 0~ 10 0 μg/ml)、潘生丁 5 μg/ml能够增加MDR细胞内阿霉素的浓度 ,且上述作用随浓度的增加而增强 ,其中红霉素 10 0 μg/ml作用最强 ,潘生丁 1μg/ml、P gp单克隆抗体、复方丹参无明显影响。结论 mdrl基因及其P gp蛋白的表达与胶质瘤细胞产生的MDR有关 ;异搏定、红霉素和潘生丁对胶质瘤细胞MDR有逆转作用

【Abstract】 Objective To investigate the mechanism of multidrug resistance (MDR) in human glioma cell and the way to overcome MDR. Methods The MDR cell line C6/adr was established.The expression of mdr 1 gene and its P glyco protein(P gp) in the C6/adr cell lines were observed by RT PCR and immunohistochemical staining. And the reversal of MDR by verapamil, erythromycin, dihydropyridine, P gp monclonal antibody, and dansen were studied with MTT assay or HPLCA.Results Mdr 1 gene of C6/adr cell line was positive, which overexpressed P gp. The drug resistance of C6/adr cell line coule be partly reversed by 2~6μg/ml of verapamil or 50~100μg/ml of erythromycin, 5μg/ml of dihydropyridine. As concentration in crease, they had better results. Among these drugs, 100μg/ml of erythromycin. Among these drugs, 100μg/ml of erythromycin had the best results.1μg/ml of dihydrpyridine, P gp monoclone antibody, dansen had no effect.Conclusions Mdr l gene and its D gp expression might be in association with the MDR of glioma cells.Verapamil, erythromycin and dihydropyridine could reverse MDR of glioma cells.
