增塑剂及热处理对水性包衣美沙拉秦结肠靶向微丸体外释药的影响

【摘要】 以乙基纤维素水分散体 Aquacoat为包衣材料 ,制备延迟释药的美沙拉秦结肠靶向微丸 ,考察了 4种增塑剂(三乙酸甘油酯、癸二酸二丁酯、枸橼酸三乙酯、邻苯二甲酸二乙酯 )和热处理时间对微丸释药的影响。结果显示 ,以枸橼酸三乙酯塑化微丸的释药最为理想 ;在延迟 3~ 4 h后开始释药 ,12 h内释药完全 ,于 60°C处理 4 h后 ,释药即趋于稳定。

【Abstract】 Mesalazine (5 ASA) delayed release pellets for colon targeted delivery were prepared with aqueous ethylcellulose dispersion(Aquacoat ECD 30) containing different plasticizers——triacetin(TRI),dibutyl sebacate(DBS),triethyl citrate(TEC),or diethyl phthalate(DEP) as coating material.The effects of plasticizer types and curing time on drug release were studied.Results indicated that the release from coated pellets containing 25% TEC or DEP was initiated after a lag time of 3~4 h,and completed within 12 h,and a constant release pattern was approached after a 4 h curing period.It is therefore consi dered that TEC is a suitable plasticizer for mesalazine colon targeted pellets coated with Aquacoat.
