纤溶酶原激活物抑制物-1在糖尿病大鼠肾皮质的表达和牛磺酸的调节作用

【摘要】 目的 研究纤溶酶原激活物抑制物 1(PAI 1)在糖尿病大鼠肾皮质的表达和牛磺酸对它的调节作用。方法 链脲佐菌素诱发的糖尿病大鼠接受牛磺酸治疗 14周后 ,测定肾皮质血管紧张素转换酶 (ACE)活性、血管紧张素Ⅱ (AngⅡ )含量、组织型纤溶酶原激活物 (t PA)和PAI 1生物活性。用Northern杂交检测肾皮质PAI 1mRNA表达。结果 与对照组大鼠相比 ,糖尿病组大鼠肾皮质ACE、PAI 1活性和AngⅡ含量均明显升高 ,t PA活性下降 ,肾皮质PAI 1mRNA表达显著增加。糖尿病大鼠用牛磺酸治疗后减少肾皮质ACE、PAI 1活性和AngⅡ含量 ,增加t PA活性 ,降低肾皮质PAI 1mRNA表达。结论 糖尿病大鼠肾皮质PAI 1基因表达上调 ,这可能与肾小球细胞外基质积聚有一定联系。牛磺酸对PAI 1基因表达的增加有抑制作用。

【Abstract】 Objective To investigate the expression of plasminogen activator inhibitor 1(PAI 1) in renal cortex of streptozotocin induced diabetic rats and its modulation by taurine Methods After taurine was given to streptozotocin diabetic rats for 14 weeks, angiotensin converting enzyme (ACE) activity, angiotensin Ⅱ(AngⅡ) content, tissue type plasminogen activator (t PA) and PAI 1 bioactivity in renal cortex were determined PAI 1 mRNA expression in renal cortex was measured by Northern blot hybridization Results Diabetic rats had an increase in ACE, PAI 1 activities, Ang Ⅱ content and PAI 1 mRNA expression, but a decrease in t PA bioactivity in the renal cortex as compared with the control animals Administration of taurine to diabetic rats reduced ACE, PAI 1 activities, Ang Ⅱ content and PAI 1 mRNA expression, but increased t PA bioactivity in the renal cortex Inhibition of PAI 1 mRNA expression was found in the renal cortex of taurine treated diabetic rats Conclusion PAI 1 mRNA expression in renal cortex of diabetic rats is upregulated This is probably related to glomerular extracellular matrix accumulation Taurine can suppress the increased expression of PAI 1 in renal cortex
