桥本甲状腺炎患者甲状腺与外周血中单个核细胞免疫功能比较
Comparison of immune function of mononuclear cells from thyroid and peripheral blood in Hashimoto’s thyroiditis
【摘要】 目的 研究桥本甲状腺炎 (HT)患者甲状腺内单个核细胞 (TMC)和外周血中单个核细胞 (PBMC)产生自身抗体和白细胞介素 1(IL 1)、TNFα的能力。方法 甲状腺球蛋白抗体 (TgAb)和抗甲状腺过氧化物酶抗体 (TPOAb)采用放射免疫测定法 ,IL 1和TNFα采用生物学测定法。结果(1)HT患者TMC和PBMC均能产生TgAb ,但TMC能力强于PMBC(P <0 .0 1) ,TMC能产生TPOAb ,而PBMC则不能 ;(2 )HT患者TMC、PBMC均可产生IL 1和TNFα ,但PBMC的产生能力明显弱于TMC(P <0 .0 1) ;(3)HT患者血清中TgAb、TPOAb分别与TMC产生的TgAb、TPOAb呈正相关 (P <0 .0 5 ) ,且血清中的TPOAb水平还与TMC产生的TNFα呈正相关 (P <0 .0 5 )。结论 甲状腺局部产生自身抗体和细胞因子所致的免疫紊乱可能是引起HT的重要原因
【Abstract】 Objective To investigate the ability to produce autoantibodies and cytokines of mononuclear cells from thyroid tissue (TMC) and peripheral blood (PBMC) in Hashimoto’s disease. Methods Antithyroglobulin antibody (TgAb) and antithyroperoxidase antibody (TPOAb) were detected by radioimmunoassay. Interleukin 1 (IL 1) and tumor necrosis factor α (TNF α) were assayed by biological detection. Results (1)TgAb was produced by TMC and PBMC, and the producing ability by TMC was significantly higher than that by PBMC. TMC synthesize TPOAb, but PBMC did not. (2)TMC and PBMC secreted IL 1 and TNFα in vitro, but the secretions of two cytokines were markedly lower in PBMC. (3)Positive correlations existed between serum TgAb (TPOAb) and TgAb (TPOAb) from TMC of HT patients. Beside, a positive correlation was also revealed between serum TPOAb and TNFα produced by TMC. Conclusion Immune disorder of thyroid seems the main cause of Hashimoto’s thyroiditis.
