异丙酚对小鼠应激溃疡后下丘脑c-fos基因表达的影响
Effects of propofol on c-fos gene expression in the hypothalamus after water-immersion induced gastric stress ulcer in mice
【摘要】 目的 检测浸水应激鼠下丘脑c fos基因的表达及异丙酚对其的影响。方法 2 4只昆明纯种小白鼠分成 3组 ,A组为正常对照组 ,B组为浸水组 ,C组为给药浸水组 ,其中C组分 2个亚组 ,C1组是在浸水后 10min腹腔注射异丙酚 3mg/kg ,C2组是腹腔注射异丙酚 3mg/kg后在行浸水。采用S P免疫组化检测方法计数各组鼠下丘脑c fos阳性细胞数。采用Guth法计数胃溃疡指数。结果 胃粘膜A组小鼠基本正常 ,B组可见明显的出血点 ,溃疡指数 6 2± 2 1,C1组和C2组溃疡指数分别为 3 2± 1 0和 2 1± 0 9,与B组比较 ,明显减少 (P <0 0 1)。下丘脑c fos基因阳性细胞百分率A组为 0 % ,B组明显增加致 6 4 2 %± 2 2 % ,C1组和C2组阳性细胞百分率分别为 2 1 8%± 3 2 %和 18 4%± 3 5 % ,均明显低于B组 (P <0 0 1)。结论 浸水应激可导致胃粘膜损伤 ,同时诱导下丘脑c fos基因表达。异丙酚可抑制其c fos基因表达 ,并减轻胃粘膜的损伤
【Abstract】 Objective To detect the c fos proto oncogene expression in the hypothalamus of the mice which underwent water immersion and the effects of propofol Methods Twenty four mice were divided randomly into three groups:group A (normal control), group B (water immersion) and group C (water immersion+propofol) The group C was subdivided into two sub groups: in group C1 3mg/kg propofol was administered intraperitoneally 10min after water immersion; in group C2 3mg/kg propofol was given intraperitoneally 10min before water immersion The cerebral c fos positive cells were detected by S P immunohistochemical assay and the gastric ulcer index was calculated using Guth method Results Animal in group A had no gastric mucosa injury The gastric ulcer index increased significantly (6 2±2 1) with the obvious bleeding points in gastric mucosa and the rate of c fos gene positive cell increased markedly (64 2%±2 1%) in group B as compared with those in group A The gastric ulcer index and c fos gene expression rate decreased greatly in group C1 (3 2±1 0, 21 8%±3 2%) and group C2 (2 1±0 9, 18 4%±3 5%) respectively (P<0 01) Conclusions Water immersion can cause gastric mucosa injury and induce c fos gene expression in hypothalamus and propofol can inhibit effectively the c fos gene expression and relieve gastric mucosa injury in mice
