透明质酸对胚胎成纤维细胞胶原网架收缩的影响

【摘要】 目的 探讨羊水中抑制胚胎伤口收缩的成分是否为透明质酸 (HA)以及HA对胚胎伤口收缩是否有直接的抑制作用。方法 使用一种体外胚胎伤口收缩模型含成纤维细胞的胶原网架FPCL) ,观察HA浓度为 1μg/ml~ 10mg/ml时对FPCL收缩的影响。 结果 低浓度组 ( 1、5、2 0、5 0、10 0、10 0 0 μg/ml)的HA对FPCL的收缩指数 (CI)与对照组比较 ,差异无显著意义 (P >0 .0 5 ) ;高浓度组 ( 2、6、10mg/ml)的HA对FPCL的CI与对照组比较 ,差异有显著意义 (P <0 .0 5 )。结论 羊水中抑制胚胎伤口收缩的成分并非为HA ,可能胚胎伤口中高浓度的HA与羊水共同参与了胚胎伤口收缩的抑制作用。

【Abstract】 Objective To investigate if the inhibitor of fetal wound contraction in amniotic fluid is hyaluronan and if hyaluronan can inhibit fetal wound contraction directly. Methods An in vitro model of wound contraction containing the fibroblast populated collagen lattice(FPCL) was used in this study to examine the effect of hyaluronan in various concentrations of 1μg/ml~10 mg/ml on contraction of FPCL. Results No significant effect was observed in the presence of low concentrations of hyaluronan(1、5、20、50、100、1000 μg/ml) with the controlled group, while a dramatic effect was observed in the presence of high concentrations of hyaluronan(2、6、10 mg/ml).Conclusion These results showed that the component which inhibit fetal wound contraction in amniotic fluid is not hyaluronan. High concentrations of hyaluronan combined with amniotic fluid might be taken part into the inhibitation of the contraction of fetal wound.
