Wnt/Frizzled信号通路靶基因C-myc在肾癌中的生物学意义

【摘要】 目的 探讨C myc基因作为Wnt/Frizzled信号通路靶基因在肾癌生物学行为中的表达及其意义。 方法 采用细胞核蛋白印迹 (Western blot)、免疫组化及免疫细胞化学C myc蛋白半定量分析方法 ,对 32例肾癌组织及肾癌细胞株GRC I、RCC 949进行了研究。 结果 C myc在肾癌组织中阳性表达 ,细胞浆阳性率 90 .6 % (2 9/ 32 )、细胞核 75 .0 % (2 4/ 32 ) ,其表达与肾癌细胞组织分型、临床分期有较大相关性 ;C myc在GRC I中呈强阳性表达。 结论 Wnt/Frizzled信号通路及其靶基因C myc在肾癌的发生发展中具有重要作用 ,通过Wnt/Frizzled信号通路的干预 ,以C myc为靶分子的肿瘤信号转导通路治疗将有重要价值及应用前景
【Abstract】 Objective To investigate the expression and function of Wnt/Frizzled pathway target gene C myc. Methods 32 cases with renal carcinoma and renal cancer cell strains (RCC 949 and GRC I) were studied by lumi light western blotting of nuclear crude extract and immunohistochemical staining. Results The expression of C myc was positive in renal cell carcinoma, the positive rate being 90.6%(29/32) and 75.0%(24/32) in tumor cell plasma and nuclear respectively. The expression of C myc protein in GRC I cell strain was higher than RCC 949 cell strain. Expression of C myc is related to tissue type and clinical type of renal cell carcinoma. Conclusion C myc ,as a target gene of Wnt/Frizzled pathway, plays important roles in carcinogenesis of renal cancer. It will be an important and valuable target in cellular signal treatment of renal cell carcinoma.
