帕金森病大鼠神经行为和中脑酪氨酸羟化酶mRNA表达的改变
Changes of neurobehavior and expression of tyrosine hydroxylase messenger RNA in mesencephalon of rat parkinsonism model
【摘要】 目的 研究帕金森病 (PD)大鼠的神经行为学特点及其中脑酪氨酸羟化酶 (TH)mRNA表达的改变。 方法 利用阿朴吗啡诱发旋转试验和前肢功能试验 ,检测PD大鼠的神经行为学特点 ,一管法逆转录聚合酶链反应 (RT PCR)半定量检测其中脑THmRNA的表达。 结果 造模 2 8只 ,经阿朴吗啡诱发旋转试验测试成功 2 1只 ,术后第 2、第 10周 2次旋转试验的 30min旋转次数分别为2 14 6± 30 4、2 17 5± 2 6 5 ,两者差异无显著性 (P >0 0 5 )。前肢功能试验 ,PD大鼠患肢摄食颗粒数(17 71± 4 82 )比对照大鼠 (34 43± 3 5 5 )明显减少 (P <0 0 1) ;5周后第 2次测试 ,PD大鼠患肢的摄食颗粒数 (18 86± 3 89)仍明显减少 ,与对照组 (35 86± 3 78)比较差异有显著性 (P <0 0 1) ,与其第 1次测试比较没有明显改善 (P >0 0 5 )。PD大鼠中脑的THmRNA表达水平明显降低 (P <0 0 1) ,模型组THmRNA表达约为对照组的 30 44 %。 结论 帕金森病大鼠神经行为学异常表现稳定 ,中脑内THmRNA的表达下降。
【Abstract】 Objective To investigate the characteristics of neurobehavior and the expression of tyrosine hydroxylase (TH) mRNA in mesencephalon of rat parkinsonism model. Methods Turning tests induced by apomorphine and forelimb functional tests were performed. Also, the expression of TH mRNA in mesencephalon of the rats was detected by semi quantitative analysis of one step RT PCR. Results In 21 of the 28 PD rats apomorphine induced turning remained at >150 turns in 30 min, and there was no significant difference between the two turning tests at the second week 〔(214 6±30 4) turns/ 30min〕 and tenth week 〔(217 5±26 5) turns/30 min〕 of 6 OHDA treatment( P >0 05). The forelimb functional tests demonstrated that, the number of food pellets eaten by PD rats with the contralateral side of 6 OHDA lesion (17 71±4 82) was significantly reduced compared with the controls (34 43±3 55)( P <0 01); 5 weeks later, the food pellets eaten by PD rats (18 86±3 89) were not increased compared with the controls (35 86±3 78)( P <0 01). A decreased expression of TH mRNA was found in mesencephalon of PD rat by reducing to 30 44% of TH mRNA expression in mesencephalon of the controls. Conclusions This study demonstrated that the PD rat model was stable in neurobehavior and decreased expression of TH mRNA in mesencephalon.
