巴马县壮族长寿老人白细胞抗原DQ基因的多态性研究

【摘要】 目的 通过对长寿老人人类白细胞抗原 (HLA) DQA1、DQB1基因多态性的研究 ,探讨HLA与长寿的关系。 方法 应用聚合酶链反应 序列特异引物法 (PCR SSP)对巴马县 10 9名 90岁以上长寿老人、5 6名 80岁老年人及 71名健康、无血缘关系、随机抽样的当地成年人进行HLA DQA1、DQB1基因分型 ,受检者均为壮族。 结果  (1)长寿组HLA DQA1 0 10 2的频率明显高于对照组 (分别为 43 3%和 2 7 8% ,P <0 0 0 5 ) ;(2 )长寿组HLA DQA1 0 10 1及DQB1 0 30 1分别为 3 7%和 0 5 % ,明显低于对照组 15 2 %和 2 1% (P分别 <0 0 0 1及 0 0 5 ) ;(3)长寿组与 80岁组比较 ,各相应等位基因之间的频率无显著性差异。相关分析显示 ,HLA DQA1 0 10 2与长寿呈正相关 ,而DQA1 0 10 1及DQB1 0 30 1呈负相关 (OR分别为 2 0 0 2、0 2 0 77及 0 5 0 0 7,P分别 <0 0 0 5、<0 0 5及 <0 0 5 )。 结论 在巴马县的壮族人群中 ,HLA DQA1 0 10 2为长寿的有利因子 ,而DQA1 0 10 1及DQB1 0 30 1为长寿的不利因子。

【Abstract】 Objective To investigate the possible association between HLA antigens and longevity. Methods By using PCR SSP method to genotype the HLA DQA1 and DQB1 loci for 109 longeval subjects (90~103 years, mean 96 years), 56 octogenarians (80~89 years, mean 85 5 years) and 71 healthy, unrelated, randomly selected subjects (23~70 years, mean 54 6 years) who live in the vicinity. All the individuals tested were of Zhuang nationality in Bama county and their families had lived there for generations. Results The frequency of HLA DQA1 *0102 allele was significantly higher in the longeval group, as compared with that of the control group (43 3% and 27 8%, P<0 005), while the frequency of the DQA1 *0101 and DQB1 *0301 in the longeval subjects was 3 7% and 0 5%, significantly lower than that of the control subjects (15 2% and 3 7%, P<0 001 and 0 05, respectively). No change was found between the octogenarians and the longeval group. Our data suggest that HLA DQA1 *0102 may be an active factor, while the DQA1 *0101 and DQB1 *0301 may be the risk factors for human longevity(OR=2 002, 0 2077 and 0 5007, respectively). Conclusions HLA DQA1 *0102, 0101 and DQB1 *0301 may be involved in human longevity in Bama population.
