神经生长因子和脑源性神经营养因子基因转入雪旺细胞的实验研究
A study on the nerve growth factor and brain- derived neurotrophic factor genetically modified Schwann cells
【摘要】 目的 神经生长因子 (nerve growth factor, NGF)和脑源性神经营养因子( brain- derived neurotrophic factor, BDNF)是具有感觉和运动神经元营养活性的因子。为提高雪旺细胞 (Schwann cell, SC)分泌 NGF和 BDNF的能力,将 NGF和 BDNF基因转入 SC进行探讨。方法 用消化后的组织块法和差速离心法分离、纯化雪旺细胞。用脂质体转染法将 NGF和 BDNF基因导入雪旺细胞, ELISA双抗体夹心法检测 NGF和 BDNF的表达。结果 经免疫组化 S- 100法鉴定 SC纯度达 95%以上。 ELISA双抗体夹心法测定转染后不同时期的 NGF和 BDNF的浓度, 2周后表达增加, 4周后明显增加,差异有非常显著性意义 (P
【Abstract】 Objective Nerve growth factor (NGF) and brain- derived neurotrophic factor (BDNF) are effective stimulatory factors of sensory and motor neurons. Schwann cells (SC) are important components for the regeneration of the nervous system. The purpose of this study is to culture adult Schwann cells and modify them genetically by human NGF and BDNF genes to increase the formation of neurotrophic factors. Methods SCs were separated and purified with post- digestion tissue blot and time- speed difference stick. Plasmids expressing NGF and BDNF genes were transfected to SC with lipofectamine, and NGF and BDNF s levels in conditioned medium of SC were measured with enzyme- linked immunoassay sensitive assay(ELISA) in different phases of gene transfected.Results The purity of SCs was 95% by S- 100 immunohistochemistry. Compared with nontranduced Schwann cells group, the levels of NGF and BDNF began increasing after two weeks of gene transfer, and both of them were increased obviously after 4 weeks.
