创面愈合过程中两种原癌基因与碱性成纤维细胞生长因子的调控关系

【摘要】 目的 研究烧伤后皮肤两种原癌基因 (c -fos、c -myc)与碱性成纤维细胞生长因子(bFGF)基因、蛋白的表达特征 ,探讨 3种基因的调控关系 ,并以增殖细胞核抗原 (PCNA)胶原纤维含量的变化评价它们对组织修复的影响。 方法 建立深Ⅱ度烫伤大鼠模型 ,采用原位杂交、RT -PCR、免疫组化及V .G特染检测皮肤组织生长相关因子基因和蛋白表达水平的变化。 结果 烧伤皮肤组织中上述生长相关因子基因或蛋白的表达呈现一定的规律性 ,c -fos、c -myc及bFGF基因在早期活跃细胞增殖阶段分别有不同程度的表达 ,在稍后的创伤愈合阶段则表达降低 ,而胶原纤维和PCNA的含量则在修复愈合阶段开始上升。 结论 在组织修复过程中 ,c -fos、c-myc和bFGF 3种生长相关因子的基因表达存在相互的网络调控作用 ,并由此影响修复细胞的增殖与分化。在后期的愈合阶段 ,3种因子表达明显受抑制 ,可能与胶原的沉积有关。

【Abstract】 Objective To investigate the relationship between the characteristics of the proto oncogenes, c fos and c myc, and the basic fibroblast growth factor (bFGF) and the proliferating cell nuclear antigen (PCNA) in burned tissues. Methods A total of 40 Wistar rats with partial thickness burn of 30%total body surface area (TBSA) on the back were used. The expressions of c fos, c myc, bFGF and PCNA were measured with in situ hybridization, reverse transcriptase polymerase chain reaction (RT PCR), immunohistochemistry and V.G methods at 3 h, 6 h, 1 d, 3 d, 7 d and 14 d after burn. The content of collagen was also measured to evaluate the repair process. Results The expression of c fos, c myc and bFGF showed different expression models in sequence and distribution during the early stage of active cellular proliferation in the injured tissues, while the expression levels of PCNA and collagen fibers started to rise during the later stage of healing. Conclusions There exists relationship between the proto oncogenes and the growth factors in the process of early wound repair, which may have some effects on latter tissue repair.
