【摘要】 目的　从基因表达水平研究高山红景天苷对大鼠心脏内分泌功能的影响。方法　分别给各组大鼠按 15mg·kg- 1 ·d- 1 ip红景天苷氯化钠溶液。饲养 30d后 ,断头处死。冷酚法提取心房总RNA ,用 (α 32 P)标记的大鼠心钠素前体原cDNA控针与各组大鼠心房总RNA做Northernblot杂交和Dotblot杂交 ,以检测心房ANP mRNA的含量。结果　老年大鼠ANP mRNA含量明显低于青年大鼠 ,注射红景天苷后 ,老年大鼠ANP mRNA含量增加而青年大鼠减少。结论　老化过程中ANP基因表达能力明显衰退 ,红景天具有一定的抑制心脏内分泌功能老化的作用。更多还原

【Abstract】 OBJECTIVE To study the effects of gadoloside on atrial natriuretic peptide (ANP) gene expression.METHODS The rats were abdominally injected with sodium chloride solution of gadoloside 15 mg·kg -1 ·d -1 for 30 days.Atria total RNA was extracted by the cold phenol method.The ANP-mRNA contents were determined using the Northem blot and Dot blot hybridization technique with α- 32 P-Labelled r-prepro-ANP-cDNA probe.RESULTS The ANP-mRNA contents of old rats were remarkably less than that of young rats.Salidroside increased the ANP-mRNA content of old rats,whereas it decreased the ANP-mRNA content of young rats.CONCLUSION The ANP gene expression declined during aging development and salidroside possessed anti-aging effects in the heart endocrine function.更多还原
