四环素哌嗪雌酚酮对小鼠骨中c-fos、c-jun蛋白表达的影响

【摘要】 目的 考察骨定向雌激素———四环素哌嗪雌酚酮 (XW 6 30 )对骨c fos和c jun蛋白表达的影响 ,初步探讨该化合物对骨的作用机制。方法 用小鼠胚胎长骨体外培养模型考察XW6 30对骨生长的作用 ,并采用免疫组织化学方法测定其对软骨静止区、增殖区和肥大区c fos,c jun基因表达产物的影响。结果 XW6 30浓度为 10 -7mol·L-1时 ,长骨骨干和骨总长生长速度显著加快 ,在 10 -9~ 10 -7mol·L-1范围内以剂量依赖方式上调骺板各区c fos和c jun蛋白的表达。 结论 提示XW 6 30可能通过上调软骨细胞c fos和c jun表达而促进软骨内骨形成

【Abstract】 OBJECTIVE To study the effects of tetracycline piperazinylestrone (XW630), a bone targeted estrogen, on the expressions of c fos and c jun proteins in bone and to investigate the mechanism by which the compound acts on bone.METHODS The effect of XW630 on bone growth was examined in vitro by a model of cultured long bones of fetal mice. The immunohistochemical analyses of c fos and c jun proteins in resting zone, proliferative zone and hypertrophic zone were performed.RESULTS XW630 enhanced the growth of long bones at 10 -7 mol·L -1 .The levels of c fos and c jun proteins in resting zone, proliferative zone and hypertrophic zone were upregulated in a dose dependent manner.CONCLUSIONS The results suggested the possibility that XW630 enhanced endochondral bone formation by upregulating the expressions of c fos and c jun proteins in chondrocytes.
