白花前胡对野百合碱所致大鼠肺动脉高压的影响

【摘要】 目的∶研究中药白花前胡的提取物PPD对野百合碱(MCT)所致大鼠肺动脉高压的影响。方法∶用MCT制造大鼠肺动脉高压模型;以硝苯地平(Nif)为阳性对照药;PV-1聚乙烯管插管测量肺动脉压力;肺组织切片HE染色显微测量肺小动脉中膜厚度;免疫荧光法观察肺组织中Tenascin-C合成的变化。结果∶PPD可明显减少MCT引起的肺动脉压升高,而对体循环压力无明显影响;可明显减少MCT所致的右心指数的升高;可明显减轻MCT所致的肺血管损伤与肺血管重建(明显减少了炎性细胞浸润、TN-C的表达及血管平滑肌细胞的增殖)。结论∶PPD可明显抑制MCT所致大鼠肺动脉高压的形成。

【Abstract】 OBJECTIVE:To study the action of the extract of Peucedanum Praeruptorum Dunnon (PPD) on monocrotaline(MCT) induced pulmonary hypertension in rats. METHODS: MCT was used to establish the chronic pulmonary hypertension model in rats; Nifedipine was used as the positive control.The pulmonary artery pressure was monitored by R6000 polygraph system through the cannulation of PV 1 polyvinyl tube.The thickness of pulmonary artery media was measured under microscope.The composition of tenascin C in pulmonary vasculature was observed through immunofluorescence assay.RESULTS: PPD reduced the pulmonary artery pressure without significant influencing the systemic artery pressure.It decreased the right heart index and reduced the thickness of small pulmonary artery media greatly.The composition of tenascin C decreased significantly in pulmonary vasculature in rats after given PPD.CONCLUSION:PPD inhibited the MCT induced pulmonary hypertension in rats.
