骨肉瘤中血管内皮生长因子的表达及血管生成的研究

【摘要】 目的 :探讨血管内皮生长因子 (vascular endothelial growth factor ,VEGF)的表达、血管生成与骨肉瘤的关系 ,为判断骨肉瘤的恶性程度及预后提供新的指标。方法 :应用免疫组化技术分别对 46例骨肉瘤标本 (经过Price分级 )进行 VEGF和微血管密度 (m icrovessel density ,MVD)染色 ,研究 VEGF表达、微血管密度之间及其与骨肉瘤的关系。结果 :骨肉瘤标本中 Price 级与 级、 级与 级之间的 VEGF表达有显著性差异 (P <0 .0 1)。Price 、 级的 VEGF表达强度高于 Price 级的表达强度 (P <0 .0 5 )。骨肉瘤标本中 Price各级之间的 MVD有显著性差异 ,且 MVD从 Price 级到 级呈递增趋势 ,微血管密度随着 Price分级的增加而逐渐增高 (P<0 .0 5 )。VEGF表达阳性组的骨肉瘤标本 MVD明显高于 VEGF表达阴性组 ,且随着 VEGF表达的增强 ,微血管密度随之增加 (P <0 .0 5 )。结论 :分化程度低、恶性程度高的骨肉瘤 VEGF表达增强 ;骨肉瘤的分化程度越低、恶性程度越高MVD越高 ;VEGF与骨肉瘤的血管生成密切相关 ,VEGF表达增强 ,微血管密度增高 ;VEGF及 MVD可能作为判定骨肉瘤预后的参考。

【Abstract】 Objective:To investigate the relationship of vascular endothelial growth factor (VEGF) and angiogenesis to the osteosarcoma, and to provide a new predictor for judgement of malignant degree and prognosis of osteosarcoma. Methods:VEGF protein expression and microvessel density in 46 cases of human osteosarcoma were examined useing immunohistochemical staining method. MVD and VEGF expression were analyzed together with their relation to the osteosarcoma. Results:VEGF expression was significantly higher in Price Ⅱ Ⅲ osteosarcoma than that in PriceⅠosteosarcoma (P<0.05) . There was significant difference in MVD in every Price classification(P<0.05). MVD increased gradually from PriceⅠto Price Ⅲ osteosarcooma.MVD in VEGF positive tumors was significantly higher than those in VEGF negative tumors(P<0.05). MVD increased gradually with the reinforcement of VEGF expression (P<0.05). Conclusion:VEGF expression is highly related to differential degree and malignant degree in osteosarcoma; MVD is highly related to pathological degree and malignant degree in osteosarcoma; VEGF is highly related to angiogenesis of osteosarcoma; VEGF expression or MVD is one of the predictors for the differential degree and malignant degree of osteosarcoma.
