肺心病患者细胞内游离钙—钙调蛋白活性的研究
Platelet Free Calcium-calmodulin Content in Cor Pulmonale
【摘要】 目的 :通过测定肺心病患者血小板内游离钙离子浓度 ([Ca2 + ]i)和钙调蛋白 (Ca M)活性 ,探讨 [Ca2 + ]i和Ca M活性变化在肺心病、缺氧性肺动脉高压形成机制中的作用。方法 :用荧光指示剂 Fura- 2 / AM标记示踪法和磷酸二酯酶法测定 19例肺心病急性发作期患者和 2 0例健康者血小板 [Ca2 + ]i、Ca M活性。结果 :肺心病组血小板[Ca2 + ]i、Ca M活性显著高于对照组。肺心病组血小板 [Ca2 + ]i与 Ca M活性呈显著正相关 ;与动脉血氧分压 (Pa O2 )呈显著负相关 ;与动脉血二氧化碳分压 (Pa CO2 )呈显著正相关。结论 :肺心病患者 [Ca2 + ]i和 Ca M活性增高在缺氧性肺动脉高压形成和肺心病发生机制中可能具有重要作用。缺氧、二氧化碳潴留是刺激 [Ca2 + ]i增高的重要因素。
【Abstract】 Objective: To investigate the effect of platelet free calcium ([Ca 2+ ]i) and calmodulin (CaM) activity on cor pulmonale and pulmonary hypertension caused by hypoxia. Methods: We assayed the consentration of platelet [Ca 2+ ]i and the activity of calmodulin CaM respectively of 19 cor pulmonale pateints in their acute stage and of 20 healthy persons, using methods of Fura 2/AM trace labelling with fluorescence and of phosphodiesterase. Results: The concentration of [Ca 2+ ]i in cor pulmonale group is higher than that of normal group significantly. In cor pulmonale group, the activity of CaM correlated positively with [Ca 2+ ]i, negatively with arterial blood oxygen pressure (PaO 2), and positively with arterial blood carbon dioxide pressure (PaCO 2) significantly. Conclusion: The activity of [Ca 2+ ]i CaM increased in acute stage of cor pulmonale, anoxia and the retension of carbon dioxide are responsible for [Ca 2+ ]i increasing.
