固定化牛肾氨基酰化酶拆分法制备D-丙氨酸

【摘要】 研究了以中空纤维固定化牛肾氨基酰化酶拆分 DL-Ala制备 D-Ala及反应的各种优化条件和固定化酶的动力学性质。实验结果表明 ,固定化酶反应的最适 p H为 7.0 ,最适反应温度为 3 0~ 3 5℃ ;固定化酶拆分反应的酶量选择为 5mg/ ml,时空体积 SV=1 .0 ,在 p H 7.0 ,3 0℃条件下的全拆分底物浓度为 0 .1 ml/ L。固定化酶常温 1周内拆分率稳定 ,但对热不稳定 ,4 0℃条件下放置 3 0 min活力下降 3 0 %。

【Abstract】 This paper deals with the study of the resolution of DL Ala by fiber immobilized cattle kidney aminoacylase(CKA). A number of factors which have effects on the relevant activity of remaining enzyme, the resolution rate of the immobilized CKA were also studied. The results showed that the optimum pH, reaction temperature were 7.0, 30~35℃ and the optimum concentration of enzyme was 5.0 mg/ml,SV=1.0. Under these conditions, the concentration of substrate which resolutes completely was 0.1 mol/L, and the result also showed that the immobilized CKA was stable for one week under normal conditions.
