牛布氏杆菌抗独特型抗体F9小鼠免疫实验

【摘要】 本研究以牛布氏杆菌抗独特型抗体F9为免疫原免疫小鼠,对小鼠体液免疫与细胞免疫的变化以及攻毒后F9对小鼠的保护情况进行了研究。阻断ELISA试验证明所制备的F9具有与布氏杆菌S85A抗原某一决定簇相似结构的“内影像”。以F9为疫苗免疫小鼠的血清凝集试验及TANAE+细胞检测结果均证明:F9能够诱导小鼠产生布氏杆菌凝集抗体并能使小鼠外周血淋巴细胞产生对布氏杆菌S85A抗原的敏感性应答,但F9组不如布氏杆菌S19号疫苗组明显。说明F9能够刺激机体产生体液反应并能非特异性地增强细胞免疫。但攻毒试验结果又证明,F9组虽然出菌检出率在绝对数值上略小于健康对照组,但差异不显著,几乎不能对小鼠提供有效保护。

【Abstract】 The research was designed for the determination of both changes of humoral and cellular immune and protective effects on F9 attacked mice with immunization of the immunogen. Brucella abortus anti-idiotype anibody F9. Blocking ELISA test demonstrated that prepared F9 had the similar structural "inner image" to certain antigen cluster of B. abortus antigen S85 A. Serum agglutination test and T ANAE+ cell detection results proved that as a vaccine, F9 could induce B. abortus agglutinating antibody in mice and peripheral lymphocytes to respond sen- sititively to B. abortus S85 A antigen. But both of the above of F9 vaccination were not as significant as B. abortus S19. It was suggested that F9 could stimulate humoral immune and enhance cellular immune non-specifically. But attack test verified that F9 could hardly provide attacked mice effective protection, F9 group had a slightly, but non-signficantly low absolute value of bacteria detection rates.
